A study of microdialysis conducted on rats shows that modafinil reduces the quantity of GABA in posterior hypothalamus. GABA receptors are also known to supply relaxation in subjects so to some extent we can say that the vigilance after taking Modalert is due the particular effect.

Orexin/Hypocretin and Histamine Systems

Orexin is a neurotransmitter that plays a very important role in homeostatic sleep/wake regulation and circadian rhythm. It is a neuropeptide that regulates wakefulness. The neurons present in the hypothalamus inside the brain controls large activities of neurons.

In brief meaning, orexin is partially responsible for our circadian rhythm and thus has been connected with the study of narcolepsy. Neurons, when activated, elevate the dopamine-norepinephrine in areas responsible for our restiveness and increases histamine in the brain. Studies on animal have shown that flawed orexin system has to lead to a person being narcoleptic or lead its symptoms.

Likewise, histamine is an endogenous wakefulness amine and therefore histamine blockade often results in sedation. Hypothalamus and tuberomammillary nuclei are where the histamine neurons are originated through the cortex and other regions of the brain. It may be the reason why antihistamines are competent of making individuals feel drowsy. It can also control erection with the use of H2 that blocks the histamine receptors causing low testosterone uptake.
